Description

Tau aggregates are a defining hallmark of several neurodegenerative disorders collectively termed tauopathies, including Alzheimer’s disease and frontotemporal dementia, where intracellular tau inclusions are associated with progressive neuronal dysfunction and degeneration.

Tau PAD12 (0N4R) is a recombinant full-length human tau variant based on the 0N4R isoform. It contains twelve phosphomimetic mutations designed to mimic disease-associated phosphorylation events and promote the formation of Alzheimer’s disease-like paired helical filaments (PHFs) in vitro. Unlike the 0N3R isoform, 0N4R contains four microtubule-binding repeat domains (R1–R4) while still lacking the two N-terminal inserts. Its engineered phosphomimetic substitutions make it a valuable model for investigating tau filament assembly, seeding, and disease-relevant aggregation mechanisms.
