Description

Tau aggregates are a defining hallmark of several neurodegenerative disorders collectively termed tauopathies, including Alzheimer’s disease and frontotemporal dementia, where intracellular tau inclusions are associated with progressive neuronal dysfunction and degeneration.

Tau PAD12 (0N3R) is a recombinant full-length human tau variant based on the fetal 0N3R isoform. It contains twelve phosphomimetic mutations designed to mimic disease-associated phosphorylation events and promote assembly into Alzheimer’s disease-like paired helical filaments in vitro. Like fetal tau, the 0N3R PAD12 variant lacks N-terminal inserts and contains three microtubule-binding repeat domains, but its engineered phosphomimetic substitutions make it particularly useful for studying tau filament formation, seeding, and disease-relevant aggregation mechanisms.
