Description

Tau aggregates are a defining hallmark of several neurodegenerative disorders collectively termed tauopathies, including Alzheimer’s disease and frontotemporal dementia, where intracellular tau inclusions are associated with progressive neuronal dysfunction and degeneration.

Tau P301S (0N4R) is a full-length human tau isoform containing no N-terminal inserts and four microtubule-binding repeat domains. It carries the disease-associated P301S mutation within the second microtubule-binding repeat, a mutation linked to inherited forms of frontotemporal dementia. The P301S substitution increases tau’s propensity to misfold and aggregate while disrupting its normal interaction with microtubules. Due to its strong pathological relevance and aggregation-prone properties, Tau P301S (0N4R) is widely used to investigate tau fibrillization, seeding, cellular toxicity, and the molecular mechanisms underlying tau-related neurodegeneration.
