Description

Tau aggregates are a defining hallmark of several neurodegenerative disorders collectively termed tauopathies, including Alzheimer's disease and frontotemporal dementia, where intracellular tau inclusions are associated with progressive neuronal dysfunction and degeneration.

Tau P301L (0N4R) is a recombinant full-length human tau isoform containing no N-terminal inserts and four microtubule-binding repeat domains. It carries the disease-associated P301L mutation within the second microtubule-binding repeat, a mutation linked to inherited forms of frontotemporal dementia with parkinsonism linked to chromosome 17 (FTDP-17). The P301L substitution reduces tau's affinity for microtubules while markedly increasing its propensity to misfold and aggregate into pathological fibrils. Owing to its strong pathological relevance and robust aggregation behavior, Tau P301L (0N4R) is widely used to investigate tau fibrillization, seeding, cellular toxicity, and the molecular mechanisms underlying tau-mediated neurodegeneration.
