Overview
Alexotech offers a complete service regarding the manufacturing of proteins including gene synthesis, protein expression, and purification. We work closely with our customers and can in-depth design expression and purification to suit your needs. As a customer, you only have to provide the desired protein sequence. The technique is suitable both to acquire active proteins as well as proteins for immunizations. In addition, it is suitable for the preparation of uniform labeling with 15N and/or 13C isotopes suitable for NMR studies.

*Alexotech guarantees >5 mg of >95% pure protein, if this is not accomplished the cost for expression and purification is waived. If for some reason, the fusion partner cannot be removed (very rare) the protein will be delivered with its fusion partner.
Price does not include DNA synthesis and cloning but is offered for 1.49 USD/nucleotide.
Project Steps
Protein design
We begin with in-depth protein design, defining the desired properties such as structure, activity, solubility, and suitability for immunization. Alexotech’s expertise in structure-based design ensures a strategy that maximizes solubility and functionality, tailored to your project goals.
Gene synthesis and cloning
The gene of interest is synthesized de novo and cloned into an in-house expression vector. All proteins include a removable affinity tag, allowing complete removal for a fully native sequence. Fusion partners can be incorporated if needed to improve solubility.
Protein expression
Expression is tested across multiple E.coli strains to identify optimal conditions. Once established, cultures are scaled up, with the option for uniform isotope labeling using 15N and 13C.
Protein purification and removal of the fusion partner
Proteins are purified using the affinity tag. Soluble proteins are purified under native conditions, while inclusion bodies are solubilized and purified under denaturing conditions. The fusion partner is removed, yielding a high-purity, ready-to-use protein.
